What is Fludarabine？

What’s the application of Fludarabine?

• Its phosphate group is rapidly cleaved in the blood, and the remaining 2-fluoroadenosine (Fara-A) enters the cell and is phosphorylated into the active metabolite Farar-ATP to act. It is used in chronic lymphocytic leukemia. This drug is a fluorinated nucleotide analog of adenosine, which is relatively resistant to the deamidation effect of adenosine deaminase. Fludarabine phosphate is rapidly dephosphorylated to 2F-ara-A, which can be taken up by cells and then phosphorylated by intracellular deoxycytidine kinase to the active triphosphate 2F-ara-ATP. This metabolite can inhibit DNA synthesis by inhibiting nucleotide reductase, DNA polymerase (α, δ, and ε), DNA primase, and DNA ligase. In addition, it can also partially inhibit RNA polymerase II thereby reducing protein synthesis.
