Description of Bis-O-(1-methylethylidene)-D-mannitol
Bretazenil (Ro16-6028) is an imidazopyrrolobenzodiazepine anxiolytic drug that is derived from the benzodiazepine family and was invented in 1988.
It is most closely related in structure to the benzodiazepine antagonist flumazenil, although its effects are somewhat different. It is classed as a high-potency benzodiazepine due to its high-affinity binding to benzodiazepine binding sites where it acts as a partial agonist. Its profile as a partial agonist and preclinical trial data suggests that it may have a reduced adverse effect profile. In particular, bretazenil has been proposed to cause a less strong development of tolerance and withdrawal syndrome. Bretazenil differs from traditional 1,4-benzodiazepines by being a partial agonist and because it binds to α1, α2, α3, α4, α5, and α6 subunit-containing GABAA receptor benzodiazepine receptor complexes. 1,4-benzodiazepines bind only to α1, α2, α3 and α5 GABAA benzodiazepine receptor complexes.

It can be synthesized in the laboratory by the following method. A mixture of 125 g of bromododecane, 38 g of thiourea, and 250 ml of 95% ethanol is refluxed for 3 h. A solution of 30 g of sodium hydroxide and 300 ml of water is added and the mixture is refluxed for another 2 h. The fractionated oil layer is the crude thiol, and the aqueous layer is acidified and extracted with benzene. The extract was combined with crude thiol, washed with water, dried with anhydrous sodium sulfate, and distilled under reduced pressure after evaporation of benzene, and the product was collected as 165-169°C (5.19 kPa) fraction. About 80-84g was obtained with a yield of about 80%. It is used as polymerization regulator of synthetic rubber, synthetic fiber, and synthetic resin; also used in the production of PVC stabilizer, drug, insecticide, fungicide, and decontaminant; used in rubber, plastic, and pharmaceutical industry; used as a relative molecular mass regulator of polymer compound; used as a modifier of hydrophobic surface.
