Afatinib is not only useful against EGFR mutations targeted by first-generation tyrosine kinase inhibitors (TKI) such as erlotinib or geochemical bookfetinib, but also against less common mutations that are resistant to these drugs. However, it has no activity against T790M mutations that normally require third-generation drugs such as axitinib.
