What is Fludarabine ?

Fludarabine is an antimetabolic fluorinated purine nucleoside analog, which is highly selective for lymphocytes, it can inhibit the repair of DNA in resting cells and the synthesis of DNA in dividing cells, and has the effect of promoting the apoptosis of these cells, which has shown good efficacy, and is now widely used in the treatment of various hematological diseases, especially chronic B-lymphocytic leukemia.

What’s the application of Fludarabine ?

• This drug is a fluorinated nucleotide analog of adenosine that is relatively resistant to the deamidation of adenosine deaminase. Fludarabine phosphate is rapidly dephosphorylated to 2F-ara-A, which can be taken up by cells and then phosphorylated by intracellular deoxycytidine kinase to the active triphosphate 2F-ara-ATP. This metabolite inhibits DNA synthesis by inhibiting nucleotide reductase, DNA polymerase (α, δ, and ε), DNA primase, and DNA ligase. In addition, it can also partially inhibit RNA polymerase II thereby reducing protein synthesis. Its phosphate group is rapidly cleaved in the blood, and the remaining 2-fluoroadenosine (Fara-A) enters the cell and is phosphorylated into the active metabolite Farar-ATP to act. It is used in chronic lymphocytic leukemia.
